• Performance: Human monocyte derived Dendritic cells were prepared in 8 day culture. Five x 105 cells per tube were washed and incubated 45 minutes on ice with 80 ul of anti-human CD273/FITC at a 1:50 dilution (10 ug/ml). They were then washed twice and analyzed by FACS. Cells stained positive with a mean shift of 0.68 log10 fluorescent units when compared to isotype control Mouse IgG2a/FITC. Binding was blocked when reagent was pre incubated with a 10-fold excess of recombinant CD273-muIg.

• Production: Protein A purified antibody from tissue culture supernatant was reacted with FITC. Unconjugated FITC was separated from antibody/FITC conjugate by desalting column. The antibody/FITC conjugate is at 0.5 mg/ml with a Fluorescein/anitbody ratio of 6.6.
• Buffer: 50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).

Information: CD273 (B7-DC, PD-L2, Programmed Death Ligand 2) is a 25 kd member of the B7 family and is expressed mainly on a sub set of Dendritic cells. When engaged it has a stimulatory effect on the expressing DC, and delivers a down regulatory signal to cells expressing its counter receptor, CD279(PD-1). The interaction of CD273(B7-DC) and CD274(B7-H1) with their receptor, CD279(PD-1) is an important component of immune regulation. Antibody ANC8D12 binds to recombinant CD273 and cell surface CD273 expressed on monocyte derived Dendritic cells (moDC), and blocks binding of recombinant CD279 to CD273 in EIA and FACS.
