1.0 mg/ml (varies by lot)
Applicable Applications for anti-CD137L antibody
Flow Cytometry (FC/FACS), ELISA (EIA), Western Blot (WB)
Performance
Five x 105 cultured Raji cells were incubated 45 minutes on ice with 80 ul of anti-CD137L/Biotin at 5 ug/ml. Cells were washed twice and incubated with 2 degree reagent Streptavidin/R-Phycoerythrin, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.79 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20 ul of 0.5 mg/ml anti-CD137L antibody.
Conjugation
Biotin
Production
Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin), and reacted with NHS-Biotin. Unconjugated Biotin was removed from conjugate using a desalting column.
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, 5% Glycerol, 0.2% BSA, 0.04% NaN3 (as a preservative).
Immunogen
Recombinant human CD137L
Preparation and Storage
Store at 2 - 5 degree C. Freeze/Thawing is not recommended.
Product should retain activity for at least 12 months after shipping date when stored as recommended.

Related Product Information for anti-CD137L antibody
Information: Human CD137 Ligand is a type II transmembrane protein constitutively expressed by monocytes, B cells neuroblastoma cells (1), and some activated T cells. The interaction of CD137 (4-1BB) and CD137L can supply a costimulatory signal to T cells through CD137. This costimulus seems to be more effective for CD8+ effector T cells than for CD4+ helper T cells (3). The interaction can also effect a reverse signal, inducing monocyte activation (2), or T cell inactivation and programmed death (4) through CD137L. Anti-CD137L antibody clone ANC5D6 binds to CD137L present on Raji and U-937 cells, and also binds to recombinant CD137L-muCD8 in EIA. This antibody does not block the interaction between CD137 and CD137L.

Product Notes

The CD137L (Catalog #AAA666016) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The anti-human CD137L-Biotin reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's CD137L can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), ELISA (EIA), Western Blot (WB). Researchers should empirically determine the suitability of the CD137L for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CD137L, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
