Dual specificity mitogen-activated protein kinase kinase 1 is a member of the dual specificity protein kinase family, which acts as a mitogen-activated protein (MAP) kinase kinase. MAP kinases, also known as extracellular signal-regulated kinases (ERKs), act as an integration point for multiple biochemical signals.This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. As an essential component of MAP kinase signal transduction pathway, this kinase is involved in many cellular processes such as proliferation, differentiation, transcription regulation and development.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate MAP2K1 in samples. An antibody specific for MAP2K1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMAP2K1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAP2K1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAP2K1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
