Principle of the Assay||The Human IL-17E ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-17E in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human IL-17E coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-17E present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-17E antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-17E bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Interleukin-25 (IL-25), also known as interleukin-17E (IL-17E), is a protein that in humans is encoded by the IL25 gene. It is a cytokine that belongs to the IL-17 cytokine family and mapped to 14q11.2. IL-25 is secreted by type 2 helper T cells (Th2) and mast cells. It has been found that IL-25 can induce Th2-type cytokine expression and production by non-T, non-B accessory cells that express high levels of major histocompatibility complex class II and low levels of Cd11c. This cytokine also can induce NF-kappaB activation, and stimulate the production of IL8. What's more, IL-25 is an important molecule controlling immunity of the gut and has been implicated in chronic inflammation associated with the gastrointestinal tract.

NCBI Description

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants. Uniprot Description

IL25: Induces activation of NF-kappa-B and stimulates production of the proinflammatory chemokine IL-8. Proinflammatory cytokine favoring Th2-type immune responses. Belongs to the IL-17 family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human IL25 il25 (Catalog #AAA8244604) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA8244604 ELISA Kit recognizes Human IL25. AAA Biotech's IL-17E can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (EIA). Researchers should empirically determine the suitability of the IL25 il25 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IL-17E, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
