Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of IL-17E. No significant cross-reactivity or interference between IL-17E and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between IL-17E and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Sandwich
Sensitivity
1.0pg/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This IL-17E ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human IL-17E. This ELISA kit for research use only!

Principle of the Assay: IL-17E ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for IL-17E. Standards or samples are then added to the microtiter plate wells and IL-17E if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of IL-17E present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for IL-17E are added to each well to "sandwich" the IL-17E immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain IL-17E and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-17E concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Mar 2010]

Uniprot Description

IL25: Induces activation of NF-kappa-B and stimulates production of the proinflammatory chemokine IL-8. Proinflammatory cytokine favoring Th2-type immune responses. Belongs to the IL-17 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cytokine; Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 14q11.2

Cellular Component: extracellular space

Molecular Function: cytokine activity; interleukin-17E receptor binding

Biological Process: response to fungus; inflammatory response to antigenic stimulus; response to nematode; eosinophil differentiation; positive regulation of transcription from RNA polymerase II promoter

Product Notes

The Human IL 17E il25 (Catalog #AAA733818) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA733818 ELISA Kit recognizes Human IL 17E. It is sometimes possible for the material contained within the vial of "Interleukin 17E, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
