Intended Uses: The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of DNASE1L3 in human serum, plasma, tissue homogenates or other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an `antibody specific to DNASE1L3. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to DNASE1L3. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain DNASE1L3, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of DNASE1L3 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes a member of the deoxyribonuclease I family. The encoded protein hydrolyzes DNA, is not inhibited by actin, and mediates the breakdown of DNA during apoptosis. Mutations in this gene are a cause of systemic lupus erythematosus-16. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. Has DNA hydrolytic activity. Is capable of both single- and double-stranded DNA cleavage, producing DNA fragments with 3'-OH ends ().
