Description: This assay employs a two-site sandwich ELISA to quantitate IKZF2 in samples. An antibody specific for IKZF2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IKZF2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IKZF2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IKZF2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Three members of this protein family (Ikaros, Aiolos and Helios) are hematopoietic-specific transcription factors involved in the regulation of lymphocyte development. Multiple transcript variants encoding different isoforms have been found for this gene, but the biological validity of some variants has not been determined.Sequence analysis predicted that the 526-amino acid human protein, which is 97% identical to the mouse protein, contains 4 N-terminal zinc fingers that compose the DNA-binding domain, and 2 C-terminal zinc fingers that mediate protein dimerization.
