Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Uveal autoantigen with coiled-coil domains and ankyrin repeats (UACA) ELISA Kit employs a two-site sandwich ELISA to quantitate UACA in samples. An antibody specific for UACA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any UACA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for UACA is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of UACA bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Using a human sequence homologous to the bovine UACA sequence to screen a Jurkat cell cDNA library, they isolated the human homolog, which was found have the same sequence as KIAA1561. Human UACA encodes a deduced 1,449-amino acid protein with a predicted molecular mass of approximately 166 kD. It contains 6 ankyrin repeats, a leucine zipper motif, and coiled-coil domains. The bovine and human proteins share 86% sequence homology. Northern blot analysis demonstrated expression at various levels in all tissues analyzed, with highest expression in skeletal muscle.the prevalence of IgG anti-UACA autoantibodies in patients with panuveitis was significantly higher than that in healthy controls (19.6-28.1% vs 0%, p less than 0.05), indicating that autoimmunity directed against UACA is a common phenomenon in these diseases.

NCBI Description

This gene encodes a protein that contains ankyrin repeats and coiled coil domains and likely plays a role in apoptosis. Studies in rodents have implicated the encoded protein in the stimulation of apoptosis and the regulation of mammary gland involution, in which the mammary gland returns to its pre-pregnant state. This protein has also been proposed to negatively regulate apoptosis based on experiments in human cell lines in which the protein was shown to interact with PRKC apoptosis WT1 regulator protein, also known as PAR-4, and inhibit translocation of the PAR-4 receptor. Autoantibodies to this protein have been identified in human patients with panuveitis and Graves' disease. Differential expression of this gene has been observed in various human cancers.
