Intended Uses: This sandwich kit is for the accurate quantitative detection of human Tripartite Motif-containing Protein 72 (also known as TRIM72) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human TRIM72 antibody. TRIM72 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human TRIM72 Antibody is added and binds to TRIM72 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated TRIM72 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human TRIM72. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

TRIM72: Muscle-specific protein that plays a central role in cell membrane repair by nucleating the assembly of the repair machinery at injury sites. Specifically binds phosphatidylserine. Acts as a sensor of oxidation: upon membrane damage, entry of extracellular oxidative environment results in disulfide bond formation and homooligomerization at the injury site. This oligomerization acts as a nucleation site for recruitment of TRIM72-containing vesicles to the injury site, leading to membrane patch formation. Probably acts upstream of the Ca(2+)-dependent membrane resealing process. Required for transport of DYSF to sites of cell injury during repair patch formation. Regulates membrane budding and exocytosis. May be involved in the regulation of the mobility of KCNB1-containing endocytic vesicles. Belongs to the TRIM/RBCC family. 2 isoforms of the human protein are produced by alternative splicing.
