Background/Introduction: Tumor necrosis factor-Beta (TNFbeta), otherwise known as lymphotoxin-alpha (LTalpha), is a member of a family of secreted and cell surface cytokines that participate in the regulation of immune and inflammatory responses. It is highly inducible, secreted, and exists as homotrimeric molecule. TNFbeta forms heterotrimers with Lymphotoxin-beta, which anchors TNFbeta to the cell surface. It also contributes to the function of lymphatic vessels and to lymphangiogenesis during inflammation. Deficiency in TNFbeta results in a reduction in lymph flow velocity. The production of TNFbeta by lymphoid tissue inducer cells or NK cells may activate stromal cells to produce VEGF-C, a crucial mediator of lymphangiogenesis in ontogeny and inflammation and/or VEGF-A that has been implicated in lymphangiogenesis in inflammation. Furthermore, TNFbeta, which is known to induce chemokines and adhesion molecules, could contribute to the recruitment of other leukocytes, such as macrophages, that produce VEGF-C.

NCBI Description

The encoded protein, a member of the tumor necrosis factor family, is a cytokine produced by lymphocytes. The protein is highly inducible, secreted, and forms heterotrimers with lymphotoxin-beta which anchor lymphotoxin-alpha to the cell surface. This protein also mediates a large variety of inflammatory, immunostimulatory, and antiviral responses, is involved in the formation of secondary lymphoid organs during development and plays a role in apoptosis. Genetic variations in this gene are associated with susceptibility to leprosy type 4, myocardial infarction, non-Hodgkin's lymphoma, and psoriatic arthritis.
