* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.

Background: S1PR1 (sphingosine-1-phosphate receptor 1) also known as S1P1 and EDG1 (endothelial differentiation gene 1) is a protein encoded by the S1PR1 gene. S1PR1 encodes a G-protein-coupled receptor which binds the lipid signaling molecule sphingosine 1-phosphate (S1P). The protein is structurally similar to G protein-coupled receptors and is highly expressed in endothelial cells. It binds the ligand sphingosine-1-phosphate with high affinity and high specificity, and suggested to be involved in the processes that regulate the differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion.Endothelial cells, and to a lesser extent, in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin.

S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues. Highly expressed in endothelial cells and to a lesser extent, in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin. Suggested to be involved in the processes that regulate the migration/differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. S1P-induced endothelial cell migration requires AKT1-mediated phosphorylation of the third intracellular loop. Seems to be coupled to the G(i) subclass of heteromeric G proteins.
