Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Mouse S1PR1 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Mouse S1PR1 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse S1PR1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Mouse S1PR1 can be calculated.

The protein encoded by this gene is structurally similar to G protein-coupled receptors and is highly expressed in endothelial cells. It binds the ligand sphingosine-1-phosphate with high affinity and high specificity, and suggested to be involved in the processes that regulate the differentiation of endothelial cells. EDG-1: a G protein-coupled receptor for the lysosphingolipid sphingosine 1-phosphate (S1P). S1P is a bioactive lysophospholipid that elicits diverse physiological effect on most types of cells and tissues. Highly expressed in endothelial cells and to a lesser extent, in vascular smooth muscle cells, fibroblasts, melanocytes, and cells of epithelioid origin. Suggested to be involved in the processes that regulate the migration/differentiation of endothelial cells. Activation of this receptor induces cell-cell adhesion. S1P-induced endothelial cell migration requires AKT1-mediated phosphorylation of the third intracellular loop. Seems to be coupled to the G(i) subclass of heteromeric G proteins.
