Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human RHO antibody. RHO present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human RHO Antibody is added and binds to RHO in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated RHOantibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human RHO. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The protein encoded by this gene is found in rod cells in the back of the eye and is essential for vision in low-light conditions. The encoded protein binds to 11-cis retinal and is activated when light hits the retinal molecule. Defects in this gene are a cause of congenital stationary night blindness. Subsequent receptor phosphorylation mediates displacement of the bound G-protein alpha subunit by the arrestin SAG and terminates signaling (PubMed:28524165, PubMed:26200343).
