Human R-Spondin-2 (RSPO2) ELISA Kit
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: R-spondins (RSPOs), such as RSPO2, are secreted proteins that regulate beta-catenin (CTNNB1) signaling. Xenopus and human RSPO2 enhanced mouse Wnt3a signaling in transfected human embryonic kidney cells. C-terminally truncated Xenopus Rspo2, but not the full-length protein, was secreted from cells and retained its activity. Mutation analysis showed that the TSP1 domain of Xenopus Rspo2 was dispensable for activity, but the furin domains were required. RT-PCR analysis showed that Xenopus embryos injected with Wnt8 or beta-catenin induced Rspo2 and Rspo3. Overexpression of Rspo2 in Xenopus activated the Wnt/beta-catenin pathway upstream of dishevelled (DVL1), interfered with Tgfb-type growth factors, and promoted myogenesis.

These proteins are secreted ligands of leucine-rich repeat containing G protein-coupled receptors that enhance Wnt signaling through the inhibition of ubiquitin E3 ligases. A chromosomal translocation including this locus that results in the formation of a gene fusion has been identified in multiple human cancers.
