Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Purinergic Receptor P2X, Ligand Gated Ion Channel 7 (P2RX7). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Purinergic Receptor P2X, Ligand Gated Ion Channel 7 (P2RX7). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Purinergic Receptor P2X, Ligand Gated Ion Channel 7 (P2RX7), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Purinergic Receptor P2X, Ligand Gated Ion Channel 7 (P2RX7) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The product of this gene belongs to the family of purinoceptors for ATP. This receptor functions as a ligand-gated ion channel and is responsible for ATP-dependent lysis of macrophages through the formation of membrane pores permeable to large molecules. Activation of this nuclear receptor by ATP in the cytoplasm may be a mechanism by which cellular activity can be coupled to changes in gene expression. Multiple alternatively spliced variants have been identified, most of which fit nonsense-mediated decay (NMD) criteria. [provided by RefSeq, Jul 2010]
