Principle of the Assay||The Mouse CD142 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD142 in Cell Culture Supernatants, Serum, Plasma, Cell Lysates. This assay employs an antibody specific for Mouse CD142 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD142 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD142 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD142 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Tissue factor, also called platelet tissue factor, factor III, thrombokinase, or CD142 is a protein present in sub endothelial tissue and leukocytes necessary for the initiation of thrombin formation from the zymogene prothrombin. Tissue factor gene was mapped to 1pter-p21 by study of somatic cell hybrids with a species-specific sensitive chromogenic assay. This gene encodes coagulation factor III which is a cell surface glycoprotein. This factor enables cells to initiate the blood coagulation cascades, and it functions as the high-affinity receptor for the coagulation factor VII.

NCBI Description

This gene encodes a membrane-bound glycoprotein that forms the primary physiological initiator of the blood coagulation process following vascular damage. The encoded protein binds to coagulation factor VIIa and the ensuing complex catalyzes the proteolytic activation of coagulation factors IX and X. Mice lacking encoded protein die in utero resulting from massive hemorrhaging in both extraembryonic and embryonic vessels. A severe deficiency of the encoded protein in mice results in impaired uterine homeostasis, shorter life spans due to spontaneous fatal hemorrhages and cardiac fibrosis. F3: Initiates blood coagulation by forming a complex with circulating factor VII or VIIa. The [TF:VIIa] complex activates factors IX or X by specific limited protolysis. TF plays a role in normal hemostasis by initiating the cell-surface assembly and propagation of the coagulation protease cascade. Interacts with HSPE; the interaction, inhibited by heparin, promotes the generation of activated factor X and activates coagulation in the presence of activated factor VII. TF expression is highly dependent upon cell type. TF can also be induced by the inflammatory mediators interleukin 1 and TNF-alpha, as well as by endotoxin, to appear on monocytes and vascular endothelial cells as a component of cellular immune response. Lung, placenta and pancreas. Belongs to the tissue factor family. 2 isoforms of the human protein are produced by alternative splicing.
