* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Background: Members of the B class of plexins, such as PLXNB2 is related to human SEP (PLXNB1). RT-PCR detected highest PLXNB2 expression in testis and ovary, followed by placenta, lung, liver, and kidney. Much lower expression was detected in prostate and small intestine, and little to none was detected in other tissues examined. Ligand-induced dimerization of plexin B was sufficient to stimulate endogenous RhoA (ARHA) potently and induce the reorganization of the cytoskeleton in mammalian cells. Overexpression of the PDZ domain of PDZ-RhoGEF prevented cell rounding and neurite retraction in differentiated rat neural precursor cells induced by activation of endogenous PLXNB1 by semaphorin-4D.
