Preparation and Storage

The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Myosin Heavy Chain 7B, Cardiac Muscle Beta (MYH7B). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Myosin Heavy Chain 7B, Cardiac Muscle Beta (MYH7B). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Myosin Heavy Chain 7B, Cardiac Muscle Beta (MYH7B), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Myosin Heavy Chain 7B, Cardiac Muscle Beta (MYH7B) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The myosin II molecule is a multi-subunit complex consisting of two heavy chains and four light chains. This gene encodes a heavy chain of myosin II, which is a member of the motor-domain superfamily. The heavy chain includes a globular motor domain, which catalyzes ATP hydrolysis and interacts with actin, and a tail domain in which heptad repeat sequences promote dimerization by interacting to form a rod-like alpha-helical coiled coil. This heavy chain subunit is a slow-twitch myosin. Alternatively spliced transcript variants have been found, but the full-length nature of these variants is not determined.
