Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-MMP-1 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-MMP-1 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the MMP-1 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of MMP-1 can be calculated.

Background/Introduction: Matrix metalloproteinase-1 (MMP-1), also known as interstitial collagenase and fibroblast collagenase[1], is a member of the matrix metalloproteinases family. Matrix metalloproteinases are zinc-dependent proteases that degrade extracellular matrix proteins. MMP-1 is structurally formed by different proteic building blocks. The Catalytic Domain of MMP-1 is composed of five highly twisted beta-strands (sI-sV), three alpha-helix (hA-hC) and a total of eight loops, enclosing a total of five metal ions, three Ca2+ and two Zn2+, one of which with catalytic role.[2] MMP1 in the stromal-tumor microenvironment can alter the behavior of cancer cells through PAR1 to promote cell migration and invasion.[3] The biologic implications of MMP1 acting as a molecular ratchet tethered to the cell surface suggested novel mechanisms for its role in tissue remodeling and cell-matrix interaction.
