Intended Uses: The Human Leptin (LPT) test kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring leptin in biological samples of human.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the LPT present in samples reacts with the anti-LPT antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, the Detection Antibody, biotin conjugated anti-LPT, is added and complexes are formed. Following a wash step, the horseradish peroxidase (HRP) conjugated Streptavidin is added and complexes are formed. After another washing step, the complexes are assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of LPT in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of LPT in the test sample. The quantity of LPT in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

NCBI Description

This gene encodes a protein that is secreted by white adipocytes into the circulation and plays a major role in the regulation of energy homeostasis. Circulating leptin binds to the leptin receptor in the brain, which activates downstream signaling pathways that inhibit feeding and promote energy expenditure. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis, reproduction, bone formation and wound healing. Mutations in this gene and its regulatory regions cause severe obesity and morbid obesity with hypogonadism in human patients. A mutation in this gene has also been linked to type 2 diabetes mellitus development. [provided by RefSeq, Aug 2017]
