Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Leptin polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Leptin polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Leptin amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Leptin can be calculated.

Background/Introduction: Leptin is a 16 kDa protein hormone that in humans encoded by the Ob gene, the gene is located on chromosome 7 in humans. The Leptin is one of the most important adipose derived hormones. It is manufactured primarily in the adipocytes of white adipose tissue, and the level of circulating leptin is proportional to the total amount of fat in the body. It plays a critical role in the regulation of body weight by inhibiting food intake and stimulating energy expenditure. In addition to its effects on body weight, leptin has a variety of other functions, including the regulation of hematopoiesis, angiogenesis, wound healing, and the immune and inflammatory response. In the clinical, Leptin has traditionally been regarded as a link between fat mass, food intake, and energy expenditure.

NCBI Description

This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: May function as part of a signaling pathway that acts to regulate the size of the body fat depot. An increase in the level of LEP may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass.

Subunit structure: Interacts with SIGLEC6. Ref.11

Subcellular location: Secreted.

Involvement in disease: Leptin deficiency (LEPD) [MIM:614962]: A rare disease characterized by low levels of serum leptin, severe hyperphagia and intractable obesity from an early age.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.16

Sequence similarities: Belongs to the leptin family.

Product Notes

The Mouse LEP lep (Catalog #AAA355428) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355428 ELISA Kit recognizes Mouse LEP. AAA Biotech's Leptin can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of Leptin in mouse serum, plasma, body fluids, tissue lysates or cell culture supernates. Researchers should empirically determine the suitability of the LEP lep for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Leptin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
