Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Latent transforming growth factor beta binding protein 2 (LTBP2) ELISA Kit employs a two-site sandwich ELISA to quantitate LTBP2 in samples. An antibody specific for LTBP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LTBP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LTBP2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LTBP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: LTBP2 belongs to the family of latent transforming growth factor (TGF)-beta binding proteins (LTBP), which are extracellular matrix proteins with multi-domain structure. This protein is the largest member of the LTBP family possessing unique regions and with most similarity to the fibrillins. It has thus been suggested that it may have multiple functions: as a member of the TGF-beta latent complex, as a structural component of microfibrils, and a role in cell adhesion.LTBP2 has approximately 45% amino acid identity to LTBP1 and 25% identity to fibrillin-1 (FBN1) but unlike other LTBP family members does not bind to latent forms of TGFB. The C-terminal region of LTBP2 competes with LTBP1 for specific binding to the N-terminal region of FBN1, but not FBN6, in microfibrils.

The protein encoded by this gene belongs to the family of latent transforming growth factor (TGF)-beta binding proteins (LTBP), which are extracellular matrix proteins with multi-domain structure. This protein is the largest member of the LTBP family possessing unique regions and with most similarity to the fibrillins. It has thus been suggested that it may have multiple functions: as a member of the TGF-beta latent complex, as a structural component of microfibrils, and a role in cell adhesion. Uniprot Description

LTBP2: May play an integral structural role in elastic-fiber architectural organization and/or assembly. Defects in LTBP2 are the cause of primary congenital glaucoma type 3D (GLC3D). An autosomal recessive form of primary congenital glaucoma (PCG). PCG is characterized by marked increase of intraocular pressure at birth or early childhood, large ocular globes (buphthalmos) and corneal edema. It results from developmental defects of the trabecular meshwork and anterior chamber angle of the eye that prevent adequate drainage of aqueous humor. Defects in LTBP2 are the cause of microspherophakia (MCSPH). It is a rare disease characterized by smaller and more spherical lenses than normal bilaterally, an increased anteroposterior thickness of the lens and highly myopic eyes. Lens dislocation or subluxation may occur, leading to defective accommodation. Defects in LTBP2 are the cause of Weill-Marchesani syndrome 3 (WMS3). A rare connective tissue disorder characterized by short stature, brachydactyly, joint stiffness, and lens abnormalities. Belongs to the LTBP family.
