Intended Uses: This ASPA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ASPA. This ELISA kit for research use only!

Principle of the Assay: ASPA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ASPA antibody and an ASPA-HRP conjugate. The assay sample and buffer are incubated together with ASPA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ASPA concentration since ASPA from samples and ASPA-HRP conjugate compete for the anti-ASPA antibody binding site. Since the number of sites is limited, as more sites are occupied by ASPA from the sample, fewer sites are left to bind ASPA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ASPA concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the transferrin family of genes and its protein product is found in the secondary granules of neutrophils. The protein is a major iron-binding protein in milk and body secretions with an antimicrobial activity, making it an important component of the non-specific immune system. The protein demonstrates a broad spectrum of properties, including regulation of iron homeostasis, host defense against a broad range of microbial infections, anti-inflammatory activity, regulation of cellular growth and differentiation and protection against cancer development and metastasis. Antimicrobial, antiviral, antifungal and antiparasitic activity has been found for this protein and its peptides. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2014]
