Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human DNase-1 antibody. DNase-1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human DNase-1 Antibody is added and binds to DNase-1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DNase-1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human DNase-1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Serum endocuclease secreted into body fluids by a wide variety of exocrine and endocrine organs (PubMed:2251263, PubMed:11241278, PubMed:2277032). Expressed by non-hematopoietic tissues and preferentially cleaves protein-free DNA (). Among other functions, seems to be involved in cell death by apoptosis (PubMed:11241278). Binds specifically to G-actin and blocks actin polymerization (). Together with DNASE1L3, plays a key role in degrading neutrophil extracellular traps (NETs) (). NETs are mainly composed of DNA fibers and are released by neutrophils to bind pathogens during inflammation (). Degradation of intravascular NETs by DNASE1 and DNASE1L3 is required to prevent formation of clots that obstruct blood vessels and cause organ damage following inflammation ().
