Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of CYCS in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Due to the 91% homology of the sequence among different species, the kit can be used to detect human, rat and mouse samples..

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to CYCS. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to CYCS. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain CYCS, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of CYCS in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21.
