Specificity
This assay has high sensitivity and excellent specificity for detection of CLDN-15. No significant cross-reactivity or interference between CLDN-15 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CLDN-15 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CLDN-15 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of HumanCLDN-15. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CLDN-15 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CLDN-15 antibody and an CLDN-15-HRP conjugate. The assay sample and buffer are incubated together with CLDN-15-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLDN-15 concentration since CLDN-15 from samples and CLDN-15-HRP conjugate compete for the anti-CLDN-15 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLDN-15 from the sample, fewer sites are left to bind CLDN-15-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN-15 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets, and also play critical roles in maintaining cell polarity and signal transductions. Alternatively spliced transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jun 2010]

Uniprot Description

Claudin-15: Plays a major role in tight junction-specific obliteration of the intercellular space, through calcium- independent cell-adhesion activity. Belongs to the claudin family.

Protein type: Cell adhesion; Membrane protein, integral; Membrane protein, multi-pass

Product Notes

The Human CLDN15 cldn15 (Catalog #AAA7231637) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7231637 ELISA Kit recognizes Human CLDN15. It is sometimes possible for the material contained within the vial of "Claudin 15 (CLDN15), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
