Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CLDN12 antibody. CLDN12 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CLDN12 Antibody is added and binds to CLDN12 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CLDN12 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CLDN12. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets, and also play critical roles in maintaining cell polarity and signal transductions. The protein encoded by this gene is a developmentally expressed renal tight junction protein. This gene is expressed in the cecum, colon, liver and kidney of mice, but is not identified in rat tissues. Humans and chimpanzees lack this gene. [provided by RefSeq, Aug 2010]
