This assay has high sensitivity and excellent specificity for detection of CD180. No significant cross-reactivity or interference between CD180 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CD180 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CD180 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CD180. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CD180 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CD180 antibody and an CD180-HRP conjugate. The assay sample and buffer are incubated together with CD180-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD180 concentration since CD180 from samples and CD180-HRP conjugate compete for the anti-CD180 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD180 from the sample, fewer sites are left to bind CD180-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD180 concentration in each sample is interpolated from this standard curve.

NCBI Description

CD180 is a cell surface molecule consisting of extracellular leucine-rich repeats (LRR) and a short cytoplasmic tail. The extracellular LRR is associated with a molecule called MD-1 and form the cell surface receptor complex, RP105/MD-1. It belongs to the family of pathogen receptors, Toll-like receptors (TLR). RP105/MD1, by working in concert with TLR4, controls B cell recognition and signaling of lipopolysaccharide (LPS), a membrane constituent of Gram-negative bacteria. [provided by RefSeq, Jul 2008]
