Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for beta NGF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any beta NGF present is bound by the immobilized antibody. Following incubation unbound samples are removed during a wash step, and then a detection antibody specific for beta NGF is added to the wells and binds to the combination of capture antibody-beta NGF in sample. Following a wash to remove any unbound combination, and enzyme conjugate is added to the wells. Following incubation and wash steps, a substrate is added. A colored product TMB is formed in proportion to the amount of beta NGF present in the sample. The reaction is terminated by addition of acid and absorbance is measured. A standard curve is prepared from seven beta NGF standard dilutions and beta NGF sample concentration determined.!!Background/Introduction: Nerve growth factor (beta NGF) is a secreted growth factor, vital for the survival, growth and maintenance of several populations of neurons in the central and peripheral nervous systems. Altered concentrations of beta NGF are also implicated in numerous neurodegenerative diseases. For instance, increased BETA NGF concentrations in serum have been reported in patients suffering from Huntington's disease and there is a loss of beta NGF in brain in patients with identified Alzheimer's disease. Further, administrating beta NGF in clinical trials has shown positive results in diabetic polyneuropathy, HIV associated peripheral neuropathy, Alzheimer's disease and skin ulcers.
