Principle of the Assay: The LumiAb Human NAP-2 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human NAP-2 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human NAP-2 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human NAP-2 or Neutrophil-Activating Factor 2, mainly known as Platelet Basic Protein, is a 128 amino acid cytokine protein encoded by the PPBP gene located at locus 4q12-4q13 on chromosome 4. After initial synthesis, the 34 residue signal sequence is cleaved from the protein, allowing the 94 residue NAP-2 protein to mature and fold properly. A series of post-translational modifications can take place to produce a variety of other factors. Proteolytic removal of residues 1-9 produces the active peptide connective tissue-activating peptide III (CTAP-III) (low-affinity platelet factor IV (LA-PF4)) while proteolytic removal of residues 1-13 produces the active peptide beta-thromboglobulin, which is released from platelets along with platelet factor 4 and platelet-derived growth factor. NAP-2 (1-66) is produced by proteolytic processing, probably after secretion by leukocytes other than neutrophils. NAP-2 (73) and NAP-2 (74) seem to not be produced by proteolytic processing of secreted precursors, but are released in an active form from platelets. LA-PF4 stimulates DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and synthesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by human synovial cells. NAP-2 is a ligand for CXCR1 and CXCR2, and NAP-2, NAP-2 (73), NAP-2 (74), NAP-2 (1-66), and most potent NAP-2 (1-63) are chemoattractants and activators for neutrophils. TC-1 and TC-2 are antibacterial proteins, in vitro released from activated platelet alpha-granules. CTAP-III (1-81) is more potent than CTAP-III desensitize chemokine-induced neutrophil activation.
