Principle of the Assay: The LumiAb Human IL-6 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-6 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-6 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-6 or Interleukin-6, also known as B-Cell Stimulatory Factor 2, is a 212 amino acid cytokine protein with a wide variety of biological functions. Expressed from the IL6 gene, the IL-6 protein is a potent inducer of the acute phase response, plays an essential role in the final differentiation of B-cells into Ig-secreting cells involved in lymphocyte and monocyte differentiation, induces myeloma and plasmacytoma growth and induces nerve cells differentiation. Typically, IL-6 is known to act on B-cells, T-cells, hepatocytes, hematopoietic progenitor cells and cells of the CNS. Also acting as a myokine, IL-6 is discharged into the bloodstream after muscle contraction and acts to increase the breakdown of fats and to improve insulin resistance. In addition, the IL-6 cytokine has been shown to be an endogenous pyrogen capable of inducing fever in people with autoimmune diseases or infections. The protein is primarily produced at sites of acute and chronic inflammation, where it is secreted into the serum and induces a transcriptional inflammatory response through interleukin-6 receptor, alpha. The functioning of this gene is implicated in a wide variety of inflammation-associated disease states, including susceptibility to diabetes mellitus and systemic juvenile rheumatoid arthritis.
