This is a TMB colorimetric sandwich ELISA kit with short assay time and quick experiment set up.
Principle
Human CXCL10 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human CXCL10 in cell culture supernatants, serum, plasma (heparin, EDTA), saliva and urine. It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Human CXCL10 with immunogen: Expression system for standard: E Coli; Immunogen sequence: V22-P98.

To measure Human CXCL10, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human CXCL10 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CXCL10 in the sample.

Chemokine(C-X-C motif) ligand 10(CXCL10) or IP-10 is a small cytokine belonging to the CXC chemokine family that is also known as 10 kDa interferon-gamma-induced protein(gamma-IP10 or IP-10). CXCL10 is secreted by several cell types in response to IFN-gamma. These cell types include monocytes, endothelial cells and fibroblasts. CXCL10 has been attributed to several roles, such as chemoattraction for monocytes/macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis. It is a potent inhibitor of angiogenesis in vivo. The gene for CXCL10 is located on human chromosome 4 in a cluster among several other CXC chemokines. It is a RAS target gene and is overexpressed in the majority of colorectal cancers. This chemokine elicits its effects by binding to the cell surface chemokine receptor CXCR3.
