Biological Significance
IL-2, also known as T cell growth factor(TCGF), is an immunoregulatory lymphokine produced by T-cells in response to protein or mitogenic stimulation. IL-2/IL-2R signaling is required for T-cell proliferation and other fundamental functions which are essential for the immune response. IL-2 stimulates growth and differentiation of B-cells, NK cells, lymphokine activated killer cells, monocytes, macrophages and oligodendrocytes. This recombinant human IL-2 is a 15.4 kDa protein, containing 133 amino acid residues including one intra chain disulfide bond. Recombinant human IL-2 was overexpressed in E Coli and purified by conventional chromatography, after refolding of the isolated inclusion bodies in a renaturation buffer.
Bioactivity
Measured in a cell proliferation assay using CTLL2 mouse cytotoxic T cells. The ED50 for this effect is < 1.5 ng/ml.
Expression System
E Coli
Preparation and Storage
Store at 4 degree C for short term storage (1/2 weeks). Aliquot and store at -20 degree C or - 70 degree C for long term storage. Avoid repeated freeze/thaw cycles.

NCBI Description

The protein encoded by this gene is a secreted cytokine that is important for the proliferation of T and B lymphocytes. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL4) and interleukin 7 (IL7). The expression of this gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. Uniprot Description

IL2: Produced by T-cells in response to antigenic or mitogenic stimulation, this protein is required for T-cell proliferation and other activities crucial to regulation of the immune response. Can stimulate B-cells, monocytes, lymphokine- activated killer cells, natural killer cells, and glioma cells. A chromosomal aberration involving IL2 is found in a form of T-cell acute lymphoblastic leukemia (T-ALL). Translocation t(4;16)(q26;p13) with involves TNFRSF17. Belongs to the IL-2 family.

Product Notes

The IL2 il2 (Catalog #AAA836386) is an Active Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's IL2 can be used in a range of immunoassay formats including, but not limited to, SDS-PAGE. Cell line: CTLL2 (mouse cytotoxic T-cell) Maintenance Condition: 10% FBS RPMI 1640 with hIL2 Assay medium: 10% FBS RPMI 1640 without hIL2 Cell Density: 2 x 10,000 cells/well (96 well plate, final volume 100ul) Starvation: 24hr, Assay medium Incubation Time: 24hr (after sample treatment) Concentration Range: 0.039 ng/ml - 40 ng/ml Detection method: MTT assay. Researchers should empirically determine the suitability of the IL2 il2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: MPTSSSTKKT QLQLEHLLLD LQMILNGINN YKNPKLTRML TFKFYMPKKA TELKHLQCLE EELKPLEEVL NLAQSKNFHL RPRDLISNIN VIVLELKGSE TTFMCEYADE TATIVEFLNR WITFSQSIIS TLT. It is sometimes possible for the material contained within the vial of "IL2, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
