Phenomenon explanation
The possible reasons that the actual band size differs from the predicted are as follows:
1. Splice variants: Alternative splicing may create different sized proteins from the same gene.
2. Relative charge: The composition of amino acids may affects the charge of the protein.
3. Post-translational modification: Phosphorylation, glycosylation, methylation etc.
4. Post-translation cleavage: Many proteins are synthesized as pro-proteins, and then cleaved to give the active form.
5. Polymerization of the target protein: Dimerization, multimerization etc.
Usage
Reconstitute in 10mM PBS (pH7.4) to a concentration of 0.1-1.0 mg/mL. Do not vortex.
Preparation and Storage
Storage:
Avoid repeated freeze/thaw cycles.
Store at 2-8 degree C for one month.
Aliquot and store at -80 degree C for 12 months.

Stability Test:
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37 degree C for 48h, and no obvious degradation and precipitation were observed. The loss rate is less than 5% within the expiration date under appropriate storage condition.

Uniprot Description

HSD17B3: Favors the reduction of androstenedione to testosterone. Uses NADPH while the two other EDH17B enzymes use NADH. Defects in HSD17B3 are the cause of male pseudohermaphrodism with gynecomastia (MPH). These individuals have unambiguous female external genitalia at birth, but fail to menstruate at the time of expected puberty and instead virilize as evidenced by growth of the phallus. Breast development may or may not take place. Belongs to the short-chain dehydrogenases/reductases (SDR) family. 17-beta-HSD 3 subfamily.

Protein type: Lipid Metabolism - androgen and estrogen; Oxidoreductase; EC 1.1.1.64

Cellular Component: intracellular membrane-bound organelle

Molecular Function: 3-alpha(17-beta)-hydroxysteroid dehydrogenase (NAD+) activity; estradiol 17-beta-dehydrogenase activity; oxidoreductase activity; testosterone 17-beta-dehydrogenase (NADP+) activity

Biological Process: lipid metabolic process; metabolic process; steroid biosynthetic process

Product Notes

The HSD17b3 hsd17b3 (Catalog #AAA2034334) is a Recombinant Protein produced from E.coli and is intended for research purposes only. The product is available for immediate purchase. The Recombinant 17-Beta-Hydroxysteroid Dehydrogenase Type 3 (HSD17b3) reacts with Homo sapiens (Human) and may cross-react with other species as described in the data sheet. AAA Biotech's 17-Beta-Hydroxysteroid Dehydrogenase Type 3 can be used in a range of immunoassay formats including, but not limited to, Positive Control; Immunogen; SDS-PAGE; Western Blot (WB). (May be suitable for use in other assays to be determined by the end user.). Researchers should empirically determine the suitability of the HSD17b3 hsd17b3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "17-Beta-Hydroxysteroid Dehydrogenase Type 3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
