Intended Uses: This GSK3beta ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat GSK3beta. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: GSK3beta ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-GSK3beta antibody and an GSK3beta-HRP conjugate. The assay sample and buffer are incubated together with GSK3beta-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GSK3beta concentration since GSK3beta from samples and GSK3beta-HRP conjugate compete for the anti-GSK3beta antibody binding site. Since the number of sites is limited, as more sites are occupied by GSK3beta from the sample, fewer sites are left to bind GSK3beta-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GSK3beta concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a serine-threonine kinase, belonging to the glycogen synthase kinase subfamily. It is involved in energy metabolism, neuronal cell development, and body pattern formation. Polymorphisms in this gene have been implicated in modifying risk of Parkinson disease, and studies in mice show that overexpression of this gene may be relevant to the pathogenesis of Alzheimer disease. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Sep 2009]
