INTENDED USE This GPI/ALT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep GPI/ALT. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY GPI/ALT ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GPI/ALT antibody and an GPI/ALT-HRP conjugate. The assay sample and buffer are incubated together with GPI/ALT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GPI/ALT concentration since GPI/ALT from samples and GPI/ALT-HRP conjugate compete for the anti-GPI/ALT antibody binding site. Since the number of sites is limited, as more sites are occupied by GPI/ALT from the sample, fewer sites are left to bind GPI/ALT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GPI/ALT concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes cytosolic alanine aminotransaminase 1 (ALT1); also known as glutamate-pyruvate transaminase 1. This enzyme catalyzes the reversible transamination between alanine and 2-oxoglutarate to generate pyruvate and glutamate and, therefore, plays a key role in the intermediary metabolism of glucose and amino acids. Serum activity levels of this enzyme are routinely used as a biomarker of liver injury caused by drug toxicity, infection, alcohol, and steatosis. A related gene on chromosome 16 encodes a putative mitochondrial alanine aminotransaminase.[provided by RefSeq, Nov 2009]
