Principle of the Assay: GATA5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GATA5 antibody and an GATA5-HRP conjugate. The assay sample and buffer are incubated together with GATA5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GATA5 concentration since GATA5 from samples and GATA5-HRP conjugate compete for the anti-GATA5 antibody binding site. Since the number of sites is limited, as more sites are occupied by GATA5 from the sample, fewer sites are left to bind GATA5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GATA5 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a transcription factor that contains two GATA-type zinc fingers. The encoded protein is known to bind to hepatocyte nuclear factor-1alpha (HNF-1alpha), and this interaction is essential for cooperative activation of the intestinal lactase-phlorizin hydrolase promoter. May play an important role in the transcriptional program(s) that underlies smooth muscle cell diversity. Rare variants in GATA5 may be a cause of susceptibility to atrial fibrillation, a common sustained cardiac rhythm disturbance. Atrial fibrillation is characterized by disorganized atrial electrical activity and ineffective atrial contraction promoting blood stasis in the atria and reduces ventricular filling.
