Principle of the Assay: GAS6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GAS6 antibody and an GAS6-HRP conjugate. The assay sample and buffer are incubated together with GAS6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAS6 concentration since GAS6 from samples and GAS6-HRP conjugate compete for the anti-GAS6 antibody binding site. Since the number of sites is limited, as more sites are occupied by GAS6 from the sample, fewer sites are left to bind GAS6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAS6 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a gamma-carboxyglutamic acid (Gla)-containing protein thought to be involved in the stimulation of cell proliferation. This gene is frequently overexpressed in many cancers and has been implicated as an adverse prognostic marker. Elevated protein levels are additionally associated with a variety of disease states, including venous thromboembolic disease, systemic lupus erythematosus, chronic renal failure, and preeclampsia. GAS6/AXL signaling plays a role in various processes such as endothelial cell survival during acidification by preventing apoptosis, optimal cytokine signaling during human natural killer cell development, hepatic regeneration, gonadotropin-releasing hormone neuron survival and migration, platelet activation, or regulation of thrombotic responses. 4 isoforms of the human protein are produced by alternative splicing.
