Immunohistochemistry (IHC)
Quality Control
The quality of the peptide was evaluated by reversed-phase HPLC and by mass spectrometry. The peptide blocks DARPP-32 Rabbit mAb signal in immunohistochemistry.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Related Product Information for PPP1R1B enzyme
Peptide is used to block DARPP-32 Rabbit mAb reactivity in immunohistochemistry protocols.

DARPP-32 (dopamine and cyclic AMPregulated phosphoprotein, relative molecular mass 32,000) is a cytosolic protein highly enriched in medium-sized spiny neurons of the neostriatum (1). It is a bifunctional signaling molecule that controls serine/threonine kinase and serine/ threonine phosphatase activity (2). Dopamine stimulates phosphorylation of DARPP-32 through D1 receptors and activation of PKA. PKA phosphorylation of DARPP-32 at Thr34 converts it into an inhibitor of protein phosphatase 1 (1). DARPP-32 is converted into an inhibitor of PKA when phosphorylated at Thr75 by cyclin-dependent kinase 5 (CDK5) (2). Mice containing a targeted deletion of the DARPP-32 gene exhibit an altered biochemical, electrophysiological and behavioral phenotype (3).

NCBI Description

This gene encodes a bifunctional signal transduction molecule. Dopaminergic and glutamatergic receptor stimulation regulates its phosphorylation and function as a kinase or phosphatase inhibitor. As a target for dopamine, this gene may serve as a therapeutic target for neurologic and psychiatric disorders. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]

Uniprot Description

Function: Inhibitor of protein-phosphatase 1.

Subcellular location: Cytoplasm.

Post-translational modification: Dopamine- and cyclic AMP-regulated neuronal phosphoprotein.Phosphorylation of Thr-34 is required for activity

By similarity.

Sequence similarities: Belongs to the protein phosphatase inhibitor 1 family.

Product Notes

The PPP1R1B ppp1r1b (Catalog #AAA653653) is an Enzyme and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's DARPP32 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC). Researchers should empirically determine the suitability of the PPP1R1B ppp1r1b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DARPP32, Enzyme" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
