Related Product Information for anti-PPP1R1B antibody
DARPP-32 (dopamine and cyclic AMP-regulated phosphoprotein, relative molecular mass 32,000) is a cytosolic protein highly enriched in medium-sized spiny neurons of the neostriatum (1). It is a bifunctional signaling molecule that controls serine/threonine kinase and serine/threonine phosphatase activity (2). Dopamine stimulates phosphorylation of DARPP-32 through D1 receptors and activation of PKA. PKA phosphorylation of DARPP-32 at threonine 34 converts it into an inhibitor of protein phosphatase 1 (1). DARPP-32 is converted into an inhibitor of PKA when phosphorylated at threonine 75 by cyclin-dependent kinase 5 (CDK5) (2). Mice containing a targeted deletion of the DARPP-32 gene exhibit an altered biochemical, electrophysiological and behavioral phenotype (3).

NCBI Description

This gene encodes a bifunctional signal transduction molecule. Dopaminergic and glutamatergic receptor stimulation regulates its phosphorylation and function as a kinase or phosphatase inhibitor. As a target for dopamine, this gene may serve as a therapeutic target for neurologic and psychiatric disorders. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]

Uniprot Description

DARPP-32: a member of the protein phosphatase inhibitor 1 family. A dopamine- and cyclic AMP-regulated neuronal phosphoprotein. Both dopaminergic and glutamatergic (NMDA) receptor stimulation regulate the extent of DARPP32 phosphorylation, but in opposite directions. Dopamine D1 receptor stimulation enhances cAMP formation, resulting in the phosphorylation of DARPP32; phosphorylated DARPP32 is a potent protein phosphatase-1 inhibitor. NMDA receptor stimulation elevates intracellular calcium, which leads to activation of calcineurin and dephosphorylation of phospho-DARPP32, thereby reducing the phosphatase-1 inhibitory activity of DARPP32. Two alternatively-spliced isoforms have been described.

Product Notes

The PPP1R1B ppp1r1b (Catalog #AAA601945) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The DARPP32 (Dopamine and cAMP-regulated Phosphoprotein, Protein phosphatase 1 regulatory subunit 1B, PP1R1B) reacts with Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's DARPP32 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP), Immunohistochemistry (IHC), Immunofluorescence (IF). Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry and Immunofluorescence. Dilution: Western Blot: 1:1000 Immunoprecipitation: 1:50 Immunohistochemistry (Paraffin): 1:100 using citrate/TBST for antigen retrieval. Immunofluorescence (Frozen): 1:400. Researchers should empirically determine the suitability of the PPP1R1B ppp1r1b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DARPP32, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
