Materials Provided
1. Capture Antibody 1 mg/mL of rabbit anti-Human CD86/B7-2 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2ug/mL in PBS before coating.
2. Detection Antibody 0.2 mg/mL of rabbit anti-Human CD86/B7-2 monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4, store at 4 degree C). Dilute to working concentration of 0.25ug/mL in detection antibody dilution buffer before use.
3. Standard Each vial contains 42 ng of recombinant Human CD86/B7-2. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20 degree C to -80 degree C in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 200 pg/mL is recommended.

Preparation and Storage
Capture Antibody: Aliquot and store at -20 degree C to -80 degree C for up to 6 months from date of receipt. Avoid repeated freeze-thaw cycles.
Detection Antibody: Store at 4 degree C and protect it from prolonged exposure to light for up to 6 months from date of receipt. DO NOT FREEZE!
Standard: Store lyophilized standard at -20 degree C to -80 degree C for up to 6 months from date of receipt. Aliquot and store the reconstituted Standard at -80 degree C for up to 1 month. Avoid repeated freeze-thaw cycles.

CD86, also known as B-lymphocyte activation antigen B7-2 (referred to as B70), is a member of the cell surface immunoglobulin superfamily. B7-2 exists predominantly as a monomer on cell surfaces and interacts with two co-stimulatory receptors CD28 and cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) expressed on T cells, and thus induces the signal pathways which regulate T cell activation and tolerance, cytokine production, and the generation of CTL. It is indicated that contacts between B and T helper cells mediated by CD86 encourage signals for the proliferation and IgG secretion of normal B cells and B cell lymphomas. A recent study has revealed that CD86 also promotes the generation of a mature APC repertoire and promotes APC function and survival. CD86 has an important role in chronic hemodialysis, allergic pulmonary inflammation, arthritis, and antiviral responses, and thus is regarded as a promising candidate for immune therapy.
