Intended Uses: The mouse NGF ELISA has been designed for the precise analytical measurement of NGF concentrations in serum, body fluids, tissue homogenate or cell culture supernatants from mice. For research use only.

Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. An analyte-specific polyclonal antibody is pre-coated onto 96-well plates. The biotin conjugated second antibody is used as detection antibody. During the first incubation the standards and samples react with the analyte-specific pre-coated antibody. In a second incubation the biotin conjugated detection antibody completes the sandwich. After washing with wash buffer the TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiterplate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample!!Background/Introduction: Nerve growth factor (NGF) is a polypeptide involved in the regulation of growth and differentiation of sympathetic and certain sensory neurons. NGF consists of 3 types of subunits, alpha, beta and gamma, which specifically interact to form a 7S, approximately 130,000-molecular weight complex. It is mainly expressed in Sertoli and interstitial cells. The proforms of NGF are secreted and cleaved extracellularly by the serine protease plasmin and by selective matrix metalloproteinases. NGF is important for the growth, maintenance, and survival of certain target neurons (nerve cells). It also functions as a signaling molecule.

Uniprot Description

RBP4: Delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP-retinol complex interacts with transthyretin, this prevents its loss by filtration through the kidney glomeruli. Defects in RBP4 are a cause of retinol-binding protein deficiency (RBP deficiency). This condition causes night vision problems. It produces a typical 'fundus xerophthalmicus', featuring a progressed atrophy of the retinal pigment epithelium. Belongs to the calycin superfamily. Lipocalin family.
