Intended Uses: The mouse Fractalkine ELISA has been designed for the precise analytical measurement of Fractalkine concentrations in serum, plasma, body fluids, tissue homogenate or cell culture supernatants from mice. For research use only.

Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. An analyte-specific polyclonal antibody is pre-coated onto 96-well plates. The biotin conjugated second antibody is used as detection antibody. During the first incubation the standards and samples react with the analyte-specific pre-coated antibody. In a second incubation the biotin conjugated detection antibody completes the sandwich. After washing with wash buffer the TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiterplate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample!!Background/Introduction: Chemokine (C-X3-C motif) ligand 1 (CX3CL1), also known as fractalkine (in humans) and neurotactin (in mice), is a large cytokine protein of 373 amino acids, it contains multiple domains and is the only known member of the CX3C chemokine family. Its gene is located on human chromosome 16 along with some CC chemokines known as CCL17 and CCL22. CX3CL1 is produced as a long protein (with 373-amino acid in humans) with an extended mucin-like stalk and a chemokine domain on top. The mucin-like stalk permits it to bind to the surface of certain cells. However a soluble (90 kD) version of this chemokine has also been observed. CX3CL1 elicits its adhesive and migratory functions by interacting with the chemokine receptor CX3CR1.

NCBI Description

The protein encoded by this gene is a cytokine that controls the production, differentiation, and function of granulocytes and macrophages. The active form of the protein is found extracellularly as a homodimer. This gene has been localized to a cluster of related genes at chromosome region 5q31, which is known to be associated with interstitial deletions in the 5q- syndrome and acute myelogenous leukemia. Other genes in the cluster include those encoding interleukins 4, 5, and 13. [provided by RefSeq, Jul 2008]
