Application Notes
Suitable for use in Caspase 3 activity assays, studying enzyme regulation, determining target substrates, screening caspase inhibitors, or as a positive control in caspase assays. Other applications not have not been tested.
Dilution: Caspase-3 activity assay: Use 0.5-1unit per test for fluorometric caspase assay. Use 2-5 units per test for colorimetric assay.

Unit Definition:
One unit of the recombinant caspase-3 is the enzyme activity that cleaves 1nmol of the caspase substrate DEVD-pNA (pNA: pnitroanaline) per hour at 37 degree C in a reaction solution containing 50mM Hepes, pH 7.2, 50mM sodium chloride, 0.1% Chaps, 10mM EDTA, 10mM DTT, 5% glycerol.
Preparation and Storage
For long-term storage, store at -70 degree C. Lyophilized product is stable for 12 months at -70 degree C. Reconstitute with sterile PBS, 15 % glycerol. Reconstituted product is stable for 3-6 months at -70 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Caspase-3 (also know as CPP32, Yama and apopain) is a member of the interleukin-1b converting enzyme (ICE) family of cysteine proteases. Caspase-3 exists in cells as an inactive 32kD proenzyme. During apoptosis procaspase-3 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases, such as caspase-6 (Mch2), caspase-8 (Flice) and grazyme B. The processed form of caspase-3 consists of large (17 kD) and small (11 kD) subunits which associate to form the active enzyme. The active caspase-3 has been shown involving in the proteolysis of several important molecules, such as poly (ADP-ribose) polymerase (PARP), the sterol regulatory element binding proteins (SREBPs), focal adhesion kinase (FAK), and others. The active recombinant caspase-3 is routinely tested for its ability to enzymatically cleave the caspase-3 colorimetric substrate Ac-DEVD-pNA.

The active recombinant human caspase-3 was expressed in E. coli. The expressed caspase-3 spontaneously undergoes autoprocessing to yield the subunits characteristic of the native enzyme. The recombinant caspase-3 preferentially cleaves caspase-3 substrates (e.g., DEVD-AFC or DEVD-pNA). In combination with caspase-3 activity assays, the active recombinant caspase-3 is very useful in screening of caspase inhibitors for pharmaceutical applications. The recombinant enzyme can also be used as a positive control in caspase assays or in determining the specificity of caspase substrates.

NCBI Description

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 6, 7 and 9, and the protein itself is processed by caspases 8, 9 and 10. It is the predominant caspase involved in the cleavage of amyloid-beta 4A precursor protein, which is associated with neuronal death in Alzheimer's disease. Alternative splicing of this gene results in two transcript variants that encode the same protein. [provided by RefSeq, Jul 2008]

CASP3: Involved in the activation cascade of caspases responsible for apoptosis execution. At the onset of apoptosis it proteolytically cleaves poly(ADP-ribose) polymerase (PARP) at a '216-Asp-|-Gly-217' bond. Cleaves and activates sterol regulatory element binding proteins (SREBPs) between the basic helix-loop- helix leucine zipper domain and the membrane attachment domain. Cleaves and activates caspase-6, -7 and -9. Involved in the cleavage of huntingtin. Triggers cell adhesion in sympathetic neurons through RET cleavage. Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 17 kDa (p17) and a 12 kDa (p12) subunit. Interacts with BIRC6/bruce. Highly expressed in lung, spleen, heart, liver and kidney. Moderate levels in brain and skeletal muscle, and low in testis. Also found in many cell lines, highest expression in cells of the immune system. Inhibited by isatin sulfonamides. Belongs to the peptidase C14A family.
