Principle of the Assay: DPP4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DPP4 antibody and an DPP4-HRP conjugate. The assay sample and buffer are incubated together with DPP4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DPP4 concentration since DPP4 from samples and DPP4-HRP conjugate compete for the anti-DPP4 antibody binding site. Since the number of sites is limited, as more sites are occupied by DPP4 from the sample, fewer sites are left to bind DPP4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DPP4 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is identical to adenosine deaminase complexing protein-2, and to the T-cell activation antigen CD26. Acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. Its binding to CAV1 and CARD11 induces T-cell proliferation and NF- kappa-B activation in a T-cell receptor/CD3-dependent manner. Its interaction with ADA also regulates lymphocyte-epithelial cell adhesion. In association with FAP is involved in the pericellular proteolysis of the extracellular matrix (ECM), the migration and invasion of endothelial cells into the ECM. May be involved in the promotion of lymphatic endothelial cells adhesion, migration and tube formation. When overexpressed, enhanced cell proliferation, a process inhibited by GPC3. Acts also as a serine exopeptidase with a dipeptidyl peptidase activity that regulates various physiological processes by cleaving peptides in the circulation, including many chemokines, mitogenic growth factors, neuropeptides and peptide hormones.
