Related Product Information for DOT1L recombinant protein
Description: Recombinant human DOT1L (KMT4) (1-435) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.
Scientific Background: DOT1-like protein (DOT1L) specifically methylates lysine-79 of histone H3 in the globular domain. DOT1L lacks a SET-domain, which is commonly considered to mediate lysine methylation of histones (1, 2). DOT1 homologs exist in a variety of eukaryotic organisms. The activity of this methyltransferase is involved in cell cycle regulation and leukemogenesis (3).

NCBI Description

The protein encoded by this gene is a histone methyltransferase that methylates lysine-79 of histone H3. It is inactive against free core histones, but shows significant histone methyltransferase activity against nucleosomes. [provided by RefSeq, Aug 2011]

Uniprot Description

DOT1L: a protein-lysine N-methyltransferase of the seven beta-strand class. Catalyzes the mono-, di- and tri-methylation of H3K79. Methylated H3K79 is associated with active gene transcription. DOT1L activity drives proliferation of mixed lineage leukemia 1 protein (MLL)-rearranged leukemias, inducing the aberrant transcription of a number of leukemogenic genes including HOXA9 and MEIS1. Binds to DNA. Belongs to the class I-like SAM-binding methyltransferase superfamily. DOT1 family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Amino Acid Metabolism - lysine degradation; EC 2.1.1.43; Methyltransferase; Methyltransferase, protein lysine

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: intracellular membrane-bound organelle; nucleoplasm; nucleus; protein complex

Molecular Function: histone lysine N-methyltransferase activity (H3-K79 specific); histone methyltransferase activity; histone-lysine N-methyltransferase activity; protein binding

Biological Process: chromatin silencing at telomere; DNA damage checkpoint; DNA repair; positive regulation of cell proliferation; positive regulation of transcription from RNA polymerase II promoter; regulation of JAK-STAT cascade; telomere organization and biogenesis

Product Notes

The DOT1L dot1l (Catalog #AAA515487) is a Recombinant Protein produced from Sf9 insect cells using baculovirus and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 1-435. AAA Biotech's DOT1L can be used in a range of immunoassay formats including, but not limited to, Methyltransferase Assay. Researchers should empirically determine the suitability of the DOT1L dot1l for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DOT1L, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
