DnaK possesses an autophosphorylation activity and a weak 5-nucleotidase activity, cleaving the 5 phosphate groups from both ribose and deoxyribose nucleotides. The role of DnaK in ATP-dependent protein-protein interactions has been extended to normal E Coli physiology, where, like eukaryotic Hsp70 homologs, it is thought to participate in the assembly/disassembly of protein complexes.

ATP-regulated binding and release of polypeptide substrates. [More information is available at EcoGene: EG10241]. Hsc56 exhibits specificity toward Hsc62, as Hsc56 does not activate DnaK or Hsc66 ATPase activity . [More information is available at EcoCyc: EG10241].

Uniprot Description

Plays an essential role in the initiation of phage lambda DNA replication, where it acts in an ATP-dependent fashion with the DnaJ protein to release lambda O and P proteins from the preprimosomal complex. DnaK is also involved in chromosomal DNA replication, possibly through an analogous interaction with the DnaA protein. Also participates actively in the response to hyperosmotic shock.
