Specificity
This assay has high sensitivity and excellent specificity for detection of DEFA5. No significant cross-reactivity or interference between DEFA5 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between DEFA5 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This DEFA5 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human DEFA5. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DEFA5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DEFA5 antibody and an DEFA5-HRP conjugate. The assay sample and buffer are incubated together with DEFA5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DEFA5 concentration since DEFA5 from samples and DEFA5-HRP conjugate compete for the anti-DEFA5 antibody binding site. Since the number of sites is limited, as more sites are DEFA5cupied by DEFA5 from the sample, fewer sites are left to bind DEFA5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DEFA5 concentration in each sample is interpolated from this standard curve.

NCBI Description

Defensins are a family of microbicidal and cytotoxic peptides thought to be involved in host defense. They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several of the alpha defensin genes appear to be clustered on chromosome 8. The protein encoded by this gene, defensin, alpha 5, is highly expressed in the secretory granules of Paneth cells of the ileum. [provided by RefSeq, Jul 2008]

Uniprot Description

DEFA5: Has antimicrobial activity against Gram-negative and Gram-positive bacteria. Defensins are thought to kill microbes by permeabilizing their plasma membrane. All DEFA5 peptides exert antimicrobial activities, but their potency is affected by peptide processing. Belongs to the alpha-defensin family.

Product Notes

The Human DEFAlpha 5 defa5 (Catalog #AAA726865) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA726865 ELISA Kit recognizes Human DEFAlpha 5. It is sometimes possible for the material contained within the vial of "Defensin Alpha 5, Paneth Cell Specific, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
