INTENDED USE This DEF?5 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep DEF?5. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY DEF?5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-DEF?5 antibody and an DEF?5-HRP conjugate. The assay sample and buffer are incubated together with DEF?5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DEF?5 concentration since DEF?5 from samples and DEF?5-HRP conjugate compete for the anti-DEF?5 antibody binding site. Since the number of sites is limited, as more sites are occupied by DEF?5 from the sample, fewer sites are left to bind DEF?5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DEF?5 concentration in each sample is interpolated from this standard curve.

NCBI Description

Defensins are a family of microbicidal and cytotoxic peptides thought to be involved in host defense. They are abundant in the granules of neutrophils and also found in the epithelia of mucosal surfaces such as those of the intestine, respiratory tract, urinary tract, and vagina. Members of the defensin family are highly similar in protein sequence and distinguished by a conserved cysteine motif. Several of the alpha defensin genes appear to be clustered on chromosome 8. The protein encoded by this gene, defensin, alpha 5, is highly expressed in the secretory granules of Paneth cells of the ileum. [provided by RefSeq, Jul 2008]
