Intended Uses: This DDIT3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine DDIT3. This ELISA kit for research use only!

Principle of the Assay: DDIT3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DDIT3 antibody and an DDIT3-HRP conjugate. The assay sample and buffer are incubated together with DDIT3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DDIT3 concentration since DDIT3 from samples and DDIT3-HRP conjugate compete for the anti-DDIT3 antibody binding site. Since the number of sites is limited, as more sites are occupied by DDIT3 from the sample, fewer sites are left to bind DDIT3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DDIT3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the CCAAT/enhancer-binding protein (C/EBP) family of transcription factors. The protein functions as a dominant-negative inhibitor by forming heterodimers with other C/EBP members, such as C/EBP and LAP (liver activator protein), and preventing their DNA binding activity. The protein is implicated in adipogenesis and erythropoiesis, is activated by endoplasmic reticulum stress, and promotes apoptosis. Fusion of this gene and FUS on chromosome 16 or EWSR1 on chromosome 22 induced by translocation generates chimeric proteins in myxoid liposarcomas or Ewing sarcoma. Multiple alternatively spliced transcript variants encoding two isoforms with different length have been identified. [provided by RefSeq, Aug 2010]
